摘要: A kinetic colorimetric assay procedure is described for measuring lactate dehydrogenase activity, in which the reduction of NAD coupled to a tetrazolium salt, 2- p -iodophenyl-3- -nitrophenyl-5-phenyl chloride (INT), with phenazine methosulfate serving as an intermediate electron carrier. The molar absorptivity formazan INT was found be 19.3 x 103 at 503 nm, provides threefold increase sensitivity over ultraviolet (340 nm) assay. results correlate well those obtained by 340-nm methods and does not require spectrophotometer. method simple, rapid, rely on secondary enzyme standards. It requires only two reagents, are stable.
