Studies of In Vitro Activities of Voriconazole and Itraconazole against Aspergillus Hyphae Using Viability Staining

摘要: The minimal fungicidal concentrations (MFCs) of voriconazole and itraconazole for five clinical isolates each Aspergillus terreus, fumigatus, flavus, niger were determined by a broth macrodilution method. Conidial suspensions as inocula compared to hyphae since the invasive form aspergillosis is manifested appearance hyphal structures. In addition, cell viability staining with dye FUN-1 was performed assess time-dependent damage exposed various antifungal agents. With conidial MFC ranges 0.5 4 μg/ml those 0.25 2 μg/ml, whereas MFCs (2 >16 μg/ml) substantially higher (P < 0.01) both voriconazole. Only minor differences between tested antifungals observed 16 20 17 spp. appeared be killed itraconazole, respectively. results correlated closely colony counts, but time- dose-dependent levels also observed. conclusion, our study demonstrates importance type inoculum used test applicability rapid sensitive method assaying hyphae.
