摘要: The present study was designed to investigate alterations in the blastogenic response of dinitrophenyl (DNP)-modified normal human lymphocytes. Cells were isolated from heparinized blood and treated with dinitrofluorobenzene (DNFB) at ratios ranging 104 1011 molecules per cell. Unmodified DNP-modified lymphocytes cultured presence Concanavalin A, pokeweed mitogen or phytohaemagglutinin (PHA). Mixed lymphocyte cultures (MLC) set up using unmodified as both stimulators responders. cells had responses all three lectins when amounts DNFB 3 × 109 molecules. 108–4 no reduction viability determined by trypan blue exclusion, but >109 molecules, incorporation 3H-labelled amino acids, 3H-uridine 3H-thymidine decreased. MLC reactivity showed similar impairment stimulator activity dropping off following treatment responder 1010 molecules/cell. 125I-labelled antibody directed against DNP bound modified suggesting that groups may have been expressed on cell surface. Concomitantly, there changes expression HLA antigens cells, shown detection new an increase expressing previously identified antigens. Other studies, carried out parallel one, suggest these be due configurational and/or conformational membrane proteins are essential for responsiveness also define specificities. major point emerges is DNP, inferentially TNP derivitization lymphocytes, not only bring about antigenic determinants, affect intrinsic ability respond a mitogenic stimulus.
