摘要: *New tools were developed for the study of endophytic development fungal species Fusarium equiseti and Pochonia chlamydosporia in barley (Hordeum vulgare) roots. These applied to monitor host colonization patterns these potential candidates biocontrol root pathogens. * Molecular beacons specific either F. or P. designed used real-time polymerase chain reaction (PCR) quantification populations Genetic transformation isolates with green fluorescent protein (GFP) gene was carried out using an Agrobacterium tumefaciens-mediated protocol, spatial investigated by laser confocal microscopy. Quantification endophytes PCR roots gave similar results all fungi, more accurate than culturing methods. Conversely, monitoring GFP-expressing transformants showed differences behaviours two species, provided evidence a plant response against endophyte colonization. Both colonized endophytically, escaping attempts prevent growth within tissues. This strongly supports balanced antagonism between virulence colonizing defence response. Development techniques GFP will facilitate future work determine their capacity.
