摘要: The chemokine stromal cell-derived factor-1 (SDF-1) that is released by bone marrow (BM) cells and contributes to stem cell homing may also play a role in the trafficking of leukaemic cells. We analysed SDF-1-induced intracellular calcium fluxes blasts from peripheral blood patients with newly diagnosed acute myeloid leukaemia (AML) lymphoblastic (B-lineage ALL), determined effect BM cell-conditioned medium on vitro transendothelial migration (TM) measured expression SDF-1 receptor, CXCR4, flow cytometry. AML FAB M1/2 did not show TM was stimulated. In myelomonocytic (M4/5), however, induced significant increased twofold conditioned medium. M3 M4 eosinophilia (M4eo) showed intermediate activity M6 no functional activity. ALL, strong (2.5-fold) were observed. Accordingly, CXCR4 low undifferentiated (M0) AML, (M1/2) erythroid (M6) but high [mean fluorescence (MF) > 50] promyelocytic (M3) (M4/5) B-lineage ALL. conclude that, preferentially active as result differentiation-related CXCR4. Functional ALL accordance previously described early B contribute infiltration, suggested migratory response BM-derived compared circulating
