Clinical pharmacology of tamoxifen in patients with breast cancer: comparison of traditional and loading dose schedules.

摘要: Tamoxifen blood levels were measured in patients with metastatic breast cancer after single-dose and continuous administration, using a traditional dose schedule of 10 mg/m2 twice daily, single daily 20 mg/m2; several loading schedules 20-80 x 7 days, then 20-mg/m2 thereafter. Using the 10-mg/m2 schedule, values administration at time clinical responses observed greater than or equal to peak dose. 40 known be associated response present by 3 hours, as opposed days schedule. Levels obtained trough times once-daily suggest tamoxifen may safely given on basis, particularly first 8 weeks therapy.
