摘要: The serum factor inducing hemorrhagic necrosis of transplantable tumors [tumor (TNF)], and the macrophage hormone associated with cachexia in cancer certain infectious diseases [cachectin] are known to be same protein. Because an association may exist between TNF cachectic state, we wished examine effect on permeability epithelial barriers. We present data showing that affects tight junctional region cells, lowering transepithelial resistance potential difference, increasing flow solute cells across epithelium. These effects dose dependent, rapidly reversible, inhibited by a monoclonal antibody TNF-α. suggest release at various sites throughout body will cause general breakdown barrier function cell sheet. This relate observed disease states. findings similar our earlier published phorbol esters diacylglycerols junctions, suggesting protein kinase C activation involved.
