Phloretin Modulates Human Th17/Treg Cell Differentiation In Vitro via AMPK Signaling.

摘要: Context. The imbalance between T helper 17 (Th17) cell and regulatory (Treg) is involved in many immune disorders diseases. Phloretin, a dihydrochalcone structural flavonoid compound, possesses bioactive properties. However, whether phloretin could impact on the differentiation of cells not completely clear. Objective. We conducted studies to explore effect glucose uptake, proliferation, human peripheral blood CD4+ investigated mechanism inducing Th17/Treg development. Methods. Naive were purified from healthy volunteers, stimulated with anti-CD3/CD28 antibodies, polarized vitro generate Th17 or Treg cells. Glucose cycle, protein expression (phospho-Stat3, phospho-Stat5), numbers analyzed by flow cytometry. AMP-activated kinase (AMPK) signaling was western blot. Results Discussion. Phloretin inhibit uptake proliferation activated inhibition due G0/G1 phase arrest. decreased generation phospho-Stat3 as well increased phospho-Stat5 process differentiation. phosphorylation level AMPK significantly enhanced, while mTOR under treatment. inhibitor compound C (Com C) neutralize phloretin, agonist 5-aminoimidazole-4-carboxamide ribonucleotide (AICAR) balance similar during induction. Conclusion. Our results suggest that can mediate regulating metabolism via signal pathway.
