Cloning of the gamma-aminobutyric acid (GABA) rho 1 cDNA: a GABA receptor subunit highly expressed in the retina.

摘要: Abstract Type A gamma-aminobutyric acid (GABAA) receptors are a family of ligand-gated chloride channels that the major inhibitory neurotransmitter in nervous system. Molecular cloning has revealed diversity subunits compose this heterooligomeric receptor, but each previously elucidated subunit displays amino similarity conserved structural elements. We have used these highly regions to identify additional members by using polymerase chain reaction (PCR). One PCR product was isolate full-length cDNA from human retina library. The mature protein predicted sequence 458 acids long and between 30 38% identified GABAA subunits. This gene is expressed primarily transcripts also detected brain, lung, thymus. Injection Xenopus oocytes with RNA transcribed vitro produces GABA-responsive conductance expression COS cells yields GABA-displaceable muscimol binding. These features consistent our identification GABA subunit, rho 1, prominent retinal increases tissue specificity ion-channel receptor family.
