摘要: High efficiency gene transfer and expression in hepatocytes vivo can be achieved using recombinant adenoviral vectors. However, the persistence of different experimental animal models has been variable. To determine if similar differences could observed a single species, was studied inbred strains mice vector that expresses human alpha 1-antitrypsin. Marked variability ranging from several weeks (C3H/HeJ Balb/c) to more than 3 months [C57Bl/6, B10.A(2R) B10.BR] when this transduced mice. This did not correlate with H-2 type. evaluate role T B cell immunity expression, congenic C3H-scid Balb/c-scid were found have indefinite hepatocytes. These animals unlike their immunocompetent counter-parts able undergo secondary transduction vector. findings suggest as yet unidentified genetic loci influence adenovirus-mediated hepatic vivo, these effects are mediated at least part, by antigen specific immune system.
