A role for the Tgf-β/Bmp co-receptor Endoglin in the molecular oscillator that regulates the hair follicle cycle

摘要: The hair follicle is a biological oscillator that alternates growth, regression and rest phases driven by the sequential activation of proliferation/differentiation programs resident stem cell populations. niches subsequent entry into growing phase mainly regulated Wnt/β-catenin signalling, while resting are Tgf-β/Bmp/Smad activity. A major question still unresolved nature molecular switch dictates coordinated transition between both signalling pathways. Here we have focused on role Endoglin (Eng), key co-receptor for members Tgf-β/Bmp family growth factors. Using an Eng haploinsufficient mouse model report required to maintain correct cycling pattern adequate stimulation niches. We further β-catenin binds promoter depending Bmp signalling. Moreover, show interacts with Smad4 in Bmp/Eng dependent context proteins act synergistically activate transcription. These observations point existence growth/rest switching mechanism based Eng-dependent feedback crosstalk Bmp/Smad signals.
