摘要: The sequences of 34 swine H chain V-regions expressed with either IgG, IgA, or IgM C regions in adult and newborn piglets are described compared. Sixteen these were cloned by using a specific leader primer whereas 18 anchored PCR. According to the operational criterion VH family classification, i.e., > 80% DNA sequence similarity, all belong homogeneous gene family. A total 31 additional VH-bearing mu clones obtained PCR from colostrum-deprived hybridized equally probe for pan-VH probe. When consensus genes compared those humans (VHI, II, III), mouse rabbit, chicken, seem have common ancestry human VHIII family, rabbit genes, single functional chicken. peptides showed < 1% variability, deduced amino acid aa4 aa25 identical. Framework (FR)1 FR2 conserved FR3 shows greater variability. 23 30 JH identical, suggesting preferential use one JH. This frequent, putative is not similar any humans. Identical genomic Southern hybridization patterns, each 13 bands differing intensity regardless stringency, when pan-specific was used. Analyses individual strand conformational polymorphism analysis suggest that number VHIII-related genome 20.
