摘要: Abstract The collagenous extracellular matrix of bone obtained after dissociative extraction with 4 M guanidine-HCI is an optimal substratum for induction by osteogenin, a morphogenetic protein. As proteinaceous substratum, this and other collagen-based materials may be immunogenic. Thus, the search discovery non-immunogenic necessary prerequisite therapeutic application principle to skeletal repair. Bovine purified greater than 50,000-fold apparent molecular mass 28 - 42 kilodaltons, was delivered into nonresorbable porous hydroxyapatite in granular disc configuration. A total 328 preparations were bioassayed osteogenic activity subcutaneous implantation 164 Long-Evans rats. Specimens harvested at day 7,11 21 subjected alkaline phosphatase determination histologic analysis. Osteogenin combined discs induced vivo differentiation phenotype mesenchymal cells invading three-dimensional space inorganic substratum. geometry had profound influence on induction, since expression solely confined did not induce when granules identical pore dimensions. finding that biological osteogenin can restored defined insoluble form basis potential proteins.
