摘要: The integrin alpha 6 subunit pairs with both the beta 1 and 4 subunits to form a subfamily of laminin receptors. Here we report cDNA cloning primary sequence for Xenopus homologue mammalian subunit. We present data demonstrating spatial temporal expression mRNA protein during early development. Initially, transcripts are expressed in dorsal ectoderm future neural plate at end gastrulation. Later development, mRNAs variety derivatives, including developing sensory placodes (otic olfactory) commissural neurons within tube. Integrin is also elongating pronephric duct as well subset rhombencephalic crest, which will Schwann cells lining several cranial nerves (VII, VIII X). In vivo an antisense transcript animal hemisphere leads reduction expression, loss adhesion laminin, severe defects normal 35% cases, reduced levels result embryos that complete gastrulation normally but arrest neurulation prior formation plate. additional 22% develop axial defects, head or tail structures. contrast, overexpression by injection full-length has no apparent effect on embryonic Co-injection sense plasmid constructs results partial rescue antisense-generated phenotypes. These indicate critical development nervous system amphibians.
