摘要: Acridine orange test (AOT) is a simple microscopic procedure based on acid conditions to denaturant DNA followed by staining with acridine orange. The AOT measures the metachromatic shift of AO fluorescence from green (native DNA) red (denatured DNA). Orange fluoresce when it binds native and fragmented DNA. Many authors observed that ≥50% in sample normal cut-off value for fertile donors. using microscopy provides general picture status denaturation. Similarly, sperm chromatin structure assay (SCSA) cytometry SCSA Software intensity (AO) fluorescence. ratio red/red + yields percentage fragmentation, referred as fragmentation index (DFI%). semen samples less than or equal 15% DFI represent low level, greater 30% values moderate, more high levels fragmentation. In present article history, principle, mechanism, technique troubleshooting points clinical significance Test are thoroughly discussed.
