Expressed sequence tag analysis in tef (Eragrostis tef (Zucc) Trotter)

摘要: Tef (Eragrostis tef (Zucc.) Trotter) is the most important cereal crop in Ethiopia; however, there very little DNA sequence information available for this species. Expressed tags (ESTs) were generated from 4 cDNA li- braries: seedling leaf, root, and inflorescence of E. leaf Eragrostis pilosa, a wild relative tef. Clustering 3603 sequences produced 530 clusters 1890 singletons, resulting 2420 unigenes. Ap- proximately 3/4 unigenes matched protein or nucleotide public databases. Annotation associated 68% putative genes with gene ontology categories. Identification translated con- served domains revealed 389 family (Pfam), frequent which was kinase. A total 170 ESTs containing simple repeats (EST-SSRs) identified 80 EST-SSR markers developed. In addition, 19 single-nucleotide polymorphism (SNP) (or) insertion-deletion (indel) 34 intron frag- ment length (IFLP) The EST database molecular study will be valuable resources further genetic research. Resume : Le est la plus importante culture cerealiere en Ethiopie. Cependant, il existe tres peu de nucleotidiques qui soient connues pour cette espece. Des etiquettes expri- mees (EST) ont ete produites partir quatre banques d'ADNc feuilles plantule, racines plantule et inflores- cences l'E. ainsi que des plantules l'Eragrostis une espece sauvage apparentee. Une analyse groupement 3 603 permis produire contigs 1 890 singlons un 2 420 unige- nes le Environ trois quarts du etaient homologues proteiques ou nucleotidi- ques connues.
