摘要: In Dictyostelium, chemotaxis to folate during growth and cAMP aggregation is controlled via cell surface receptors. To study the role of two G alpha proteins (G 1 2) in these responses, we examined physiological biochemical effects null mutations caused by antisense mutagenesis gene disruptions. Disruption 2 results an aggregation-deficient phenotype a loss receptor-mediated functions, including activation adenylate cyclase, guanylate expression GTP-mediated decrease receptor affinity for cAMP, but it has no effect on or cyclase. These phenotypes can be rescued vector expressing 2, suggesting coupled not Loss resulted visible developmental phenotype, cAMP- folate-stimulated function either essential under standard laboratory conditions encoded multiple genes. Availability provides suitable genetic backgrounds mutant protein subunits which then used examine roles 2. Construction disruptions was facilitated using auxotrophic marker THY1, allowed selection single-copy insertions into genome.
