Desiccation-tolerant somatic embryos of Norway spruce (Picea abies) can be produced in liquid cultures and regenerated into plantlets

摘要: A rapid and efficient culture system for Picea abies somatic embryos grown in liquid has been developed. Embryogenic cultures on semisolid medium (K‐medium) supplemented with auxin (10 μM naphthaleneacetic acid) cytokinin (5 N6‐benzyladenine) 2–7 yr were used as inoculum cultures. Liquid established after 1 mo K‐medium then subcultured to a 16 abscisic acid 7.5% polyethylene glycol (LAP medium) further development of the embryos. After ca. 2 wk, large number well‐formed could be observed. Within 5 wk had initiated both cotyledons, shoot apical meristem, radicle.
