T cell repertoire expression in murine recipients of bone marrow transplant after LF 08-0299 (Tresperimus) administration

摘要: LF 08-0299 (Tresperimus), a novel immunosuppressive compound, has been previously shown to prevent graft-versus-host disease in murine models. In this study, we investigated the influence of on TCR Vbeta repertoire irradiated F1 recipient mice reconstituted with either syngeneic or parental bone marrow cells. We showed that partial blockade thymic differentiation occurred normal under treatment at transition CD4-/CD8- CD4+/CD8+, and was fully reversible. Despite effect thymus, T cell negative selection preserved following transplantation. further assessed whether administration could modify expression recipients our transplant model, abnormal Vbeta3, Vbeta5, Vbeta6 Vbeta11 demonstrated peripheral lymph nodes recipients. Moreover, Vbeta3 populations were overexpressed. Administration modified pattern expression. The expansion cells selectively inhibited therapy and, contrast, Vbeta5 Lymph node histological analysis prevented reaction occurring untreated mice.
