Mapping human brain monoamine oxidase A and B with 11C-labeled suicide inactivators and PET.

摘要: The regional distributions of monoamine oxidase (MAO) types A and B have been identified in human brain vivo with intravenously injected 11C-labeled suicide enzyme inactivators, clorgyline L-deprenyl, positron emission tomography. rapid uptake retention radioactivity for both 11C tracers indicated irreversible trapping. anatomical distribution paralleled the MAO autopsy material. corpus striatum, thalamus, brainstem contained high activity. magnitudes [11C]clorgyline L-[11C]deprenyl were markedly reduced one subject treated antidepressant inhibitor phenelzine. comparison inactive (D-) active (L-) enantiomers deprenyl showed clearance enantiomer within B-rich structures, agreement known stereoselectivity L-deprenyl. Prior treatment unlabeled L-deprenyl prevented L-[11C]deprenyl. Thus, inactivators labeled emitters can be used to quantitate kinetic characteristics structures.
