Characterization of arenaviruses using a family-specific primer set for RT-PCR amplification and RFLP analysis. Its potential use for detection of uncharacterized arenaviruses.

摘要: Arenaviruses are enveloped viruses with a genome composed of two ssRNA species, designated L and S. The arenaviruses were divided in major groups (Old World New World), based on serological properties genetic data, as well geographic distribution. A sequence alignment analysis all reported arenavirus S RNAs yielded 17 conserved regions addition to region at the end both RNAs. consensus sequences these used design generalized primers suitable for RT-PCR amplification set overlapping nucleotide fragments comprising complete RNA any arenavirus. restriction (RFLP) was designed rapidly typify amplified fragments. This RT-PCR-RFLP approach tested Old (LCM) (Junin Tacaribe) arenaviruses. Furthermore, using this procedure whole novel isolate obtained from rodent trapped central Argentina, characterized. Partial data phylogenetic analyses that showed relationships between rest members family. relatively simple methodology will be useful basic studies epidemiological survey programs.
