摘要: Metabolite fingerprinting provides a powerful method for discriminating between biological samples on the basis of differences in metabolism caused by such factors as growth conditions, developmental stage or genotype. This protocol describes technique acquiring metabolite fingerprints from plant origin. The preferred involves freezing tissue rapidly to stop metabolism, extracting soluble metabolites using perchloric acid (HClO4) and then obtaining fingerprint metabolic composition sample 1D 1H NMR spectroscopy. spectral multiple may be analyzed either unsupervised supervised multivariate statistical methods, these approaches are illustrated with data obtained developing seeds two genotypes sunflower (Helianthus annuus). Preparation extracts analysis takes 2-3 d, but can processed parallel subsequent acquisition spectra approximately 30 min per sample, allowing 24-48 week.
