摘要: Calibration of microarray measurements aims at removing systematic biases from the probe-level data to get expression estimates that linearly correlate with transcript abundance in studied samples. The improvement calibration methods is an essential prerequisite for estimating absolute levels, which, turn, are required quantitative analyses transcriptional regulation, example, context gene profiling diseases. We address hybridization on microarrays as a reaction process complex environment and express measured intensities function input quantities experiment. Popular such MAS5, dChip, RMA, gcRMA, vsn, PLIER briefly reviewed assessed light model previous benchmark studies. present our hook method, new approach based graphical summary actual characteristics particular microarray. Although single-chip related, performs well multi-chip-related presently one best state-of-the-art values. addition, provides set chip evaluate performance given hybridization. algorithm method described its exemplified.
