The primary requisite for nuclei is to isolate them in normal condition adequate amounts keep chromatin an undamaged state. Several factors control success isolation, which low temperature, medium, method homogenization, pH, divalent cations are most important should be suitably adjusted each tissue. procedure may direct or indirect.
