UVB irradiation down-regulates HPV-16 RNA expression: implications for malignant progression of transformed cells.

摘要: A new cell line obtained from normal human epithelial keratinocytes transfected with the whole HPV-16 genome has been extensively characterised. This line, named HK-168, a basal/para-basal keratinocyte phenotype, requires use of serum-free chemically defined media and maintains ability to differentiate toward pluri-stratified epithelia. Although immortalised it is not capable anchorage independent growth tumorigenic. HK-168 distinctive kariotype, complete, transcriptionally active integrated at an almost 1:1 ratio into host haploid thus providing convenient experimental model for viral transformed pre-neoplastic phenotype. The oxidative stress, induced by mild UVB irradiation, caused in general suppression transcription, accompanied moderate arrest, appropriated response cellular antioxidant enzymes, activation repair mechanisms induction apoptosis. similar one observed highly resistant diploid keratinocytes. Conversely, cells devoid HPV sequences (HaCaT), appeared extremely susceptible We propose that reported oncogenes, restoring control on mechanisms, allows damage repair, ultimately resulting surviving response.
