摘要: Factors which regulate expression of the haptoglobin (acute phase reactant) gene in adipocytes have been examined using 3T3-L1 cells. Haptoglobin was observed by Northern blotting each major white adipose tissue depots mice (epididymal, subcutaneous, mesenteric, and perirenal) interscapular brown fat. Expression occurred mature adipocytes, but not stromal-vascular fraction. In cells, mRNA detected from day 4 after induction differentiation into adipocytes. Lipopolysaccharide cytokines, TNFalpha interleukin-6, resulted substantial increases adipocytes; increase (7-fold) highest with TNFalpha. Increases level were also induced dexamethasone, noradrenaline, isoprenaline, a beta3-adrenoceptor agonist. contrast, reduced nicotinic acid PPARgamma agonist, rosiglitazone. RT-PCR showed that expressed human (subcutaneous, omental). It is concluded stimulated inflammatory glucocorticoids, sympathetic system, while activation nuclear receptor strongly inhibitory.
