摘要: Conditions were standardized for the isolation and culture of protoplasts from an embryogenic cell suspension Picea glauca. A combination 0.5% Cellulase R-10, 0.25% Macerozyme, Driselase, Rhozyme HP-150 with 0.5M mannitol 5 mM CaCl2.2H2O produced average 4.5 × 106 per gram fresh weight cells. Of several protoplast media tested, von Arnold Eriksson Kao Michayluk (KM8P) best supported mitotic divisions protoplasts. density 105 ml addition glutamine to medium was necessary induce sustained microcallus formation. Microcalli grew into subculturable callus using a nurse technique.
