摘要: Cytokines such as interleukin-6 (IL-6) and IL-17 which act key regulators of the immune response have been identified to a potential role in bone remodeling mechanism. Receptor activator NF-κB ligand (RANKL) has shown regulate osteoclast differentiation function while osteoprotegerin (OPG) blocks binding RANKL inhibits osteoclasts, thus favoring osteogenesis. Alkaline phosphatase (ALP) on other hand works early mineralization indicator regulation. The current study aims determine IL-6 IL-17A regulating OPG/RANKL system murine osteoblast cell line (MC3T3-E1). Gene expression analysis showed significant up-regulation OPG ALP by all treated groups (rIL-6, rIL-17A rIL-6 + rIL-17A). In contrast, treatment cells with and/or down-regulation expression. Interestingly, combinations rIL17A marked increased ratio. Similar pattern protein was observed osteoblasts detected western blotting ELISA. These findings suggest new mechanism regulation these cytokines RANKL, could promote osteogenesis diminish osteoclastogenesis.
