In Vitro Effects of Isoorientin, Forsythoside B, and Verbascoside on Bovine Kidney Cortex Glutathione Reductase

摘要: Glutathione reductase (GR; E.C 1.6.4.2) catalyzes the reduction of oxidized glutathione to reduced by nicotinamide adenine dinucleotide phosphate (NADPH) dependent mechanism. GR participates in protection cell from toxic effects free radicals. In this study, we have investigated vitro isoorientin (C-glycosyl flavonoid), verbascoside, and forsythoside B (phenylethanoids) on purified bovine kidney cortex GR. These compounds present many plant species that been used as medicinal plants an interesting spectrum biological activity. We demonstrated these inhibit a concentration-dependent manner. Kinetic characterization inhibition is also done. Isoorientin noncompetitive inhibitor with respect both disulfide (GSSG) (KiGSSG 0.023 ± 0.001 mM) NADPH (KiNADPH 0.032 mM). Forsythoside acts uncompetitive GSSG 0.20 0.014 0.267 0.028 Verbascoside 0.242 0.024 0.175 0.016 Inhibition causes accumulation reactive oxygen depletion pool. enzyme may be significant drug resistance.
