摘要: The hypoglycemic effect of sulfonylureas and their analogues results from binding to a high affinity site in the B-cell plasma membrane. This seems be structural component ATP-sensitive K(+)-channel represents pancreatic sulfonylurea receptor. Binding causes closure thereby initiates chain events eventually leading release insulin. Diazoxide inhibits insulin secretion via opening K(+)-channel. Sulfonylurea receptors resembling receptor occur nerve cells, cardiac muscle, skeletal muscle smooth muscle. Neither these extrapancreatic nor low for myocytes adipocytes contribute therapeutic benefit sulfonylureas.
