Neuroprotection against iron-induced cell death by perineuronal nets - an in vivo analysis of oxidative stress.

摘要: Perineuronal nets (PNs) are a specialized form of extracellular matrix, surrounding different types neurons and mainly consist chondroitin sulfate proteoglycans connected to hyaluronan, stabilized by link protein cross-linked via tenascin-R. Due their polyanionic character, caused the highly charged glycosaminoglycan hyaluronan components, PNs might be involved in local ion homeostasis. They able scavenge bind redox-active ions thus reduce oxidative potential. We investigated whether netenwrapped less vulnerable against iron-induced processes. Oxidative stress is key factor development progression neurodegenerative diseases like Alzheimer's Parkinson's disease. Iron believed contribute Alzheimer brains catalyzing generation free radicals. For examining potential neuroprotective effects PNs, mice were microinjected with 0.2μl 20mM solution FeCl3 into barrel field while control group received an equal volume 0.9% NaCl. Brains analyzed after time intervals 24h 72h. Neuronal degeneration was visualized using Fluoro-Jade B staining. The presence assessed Wisteria floribunda agglutinin histochemistry or aggrecan immunocytochemistry. analysis showed significant lower rate net-ensheathed comparison without PNs. results suggest mechanism associated cell death.
