Effects of Tumor Necrosis Factor Inhibitor on Stress-Shielded Tendons.

摘要: Mechanical stress plays an important role in preserving the integrity of bone and ligament. Stress shielding reduces mechanical load on or tendons, resulting tissue degradation. Previous studies showed that deterioration tendon structure during is associated with elevated expression tumor necrosis factor (TNF)-α. This study examined therapeutic potential TNF inhibitor etanercept preventing morphologic Achilles after shielding. Rats (N=48) were exposed to left treated phosphate-buffered saline for 2 4 weeks. The right tendons used as controls. After weeks, stress-shielded appeared less smooth than control formed adhesions surrounding tissues. Transmission electron microscopy also disarray collagen fibrils a significant increase number small-diameter fibrils. These changes increased TNF-α, matrix metalloproteinase (MMP)-13, MMP-3, I, III. Treatment weeks injection reduced organization induced by Etanercept treatment attenuated upregulation MMP-13, III levels. However, no difference was observed between group treatment. current findings show TNF-α inhibition can protect against early stages remodeling shielding, but additional interventions may be necessary prevent degeneration long-term [Orthopedics. 2017; 40(1):49-55.].
