Intravital two-photon microscopy of host-pathogen interactions in a mouse model of Staphylococcus aureus skin abscess formation.

摘要: Summary Staphylococcus (S.) aureus is a frequent cause of severe skin infections. The ability to control the infection largely dependent on rapid recruitment neutrophils (PMN). To gain more insight into dynamics PMN migration and host–pathogen interactions in vivo, we used intravital two-photon (2-P) microscopy visualize S. aureus infections mouse. Reporter strains expressing fluorescent proteins were developed, which allowed for detection bacteria vivo. By employing LysM-EGFP mice PMN, observed appearance extravascular space dermis their directed movement towards focus infection, led delineation an abscess within 1 day. Moreover, tracking transferred labelled bone-marrow showed that localization site presence G-protein-coupled receptors whereas Interleukin-1 receptor was required host cells other than PMN. Furthermore, complement inhibitor Ecb could block accumulation at thesite infection. Our results establish 2-P powerful tool investigate orchestration immune cells, location gene expression vivo single cell level.
