摘要: We have compared three different production methods, which may be suitable for the large scale of adenovirus vectors human clinical trials. The procedures 293 cells adapted to suspension growth in serum-free medium a stirred tank bioreactor, on microcarriers serum-containing and grown standard tissue culture plasticware. With given virus, yields varied between 2000 10,000 infectious units/cell. bioreactor routinely produced 4000 7000 units/cell when were microcarriers. same yielded Yields obtained from plasticware up Cell conditions monitored glucose consumption, lactate production, ammonia accumulation. Glucose consumption accumulation correlated well with cell parameters. Ammonia does not appear significant. Based virus yields, ease operation linear scalability, large-scale seems feasible using (adapted suspension/serum free or serum containing medium) bioreactor.
