摘要: Oral squamous cell carcinoma (OSCC) is a common malignancy, the incidence of which particularly high in some Asian countries due to geographically linked areca quid (AQ) chewing habit. In this study, array-based comparative genomic hybridization was used screen microdissected OSCCs for genome-wide alterations. The highest frequencies gene gain were detected TP63, Serpine1, FGF4/FGF3, c-Myc and DMD. deletion Caspase8 MTAP. Gained genes, classified by hierarchical clustering, mainly on 17q21-tel; 20q; 11q13; 3q27-29 X chromosome. Among these, gains EGFR at 7p, CCND1 EMS1 11q13, AIB1 20q significantly associated with lymph node metastasis. profiles FHIT EXT1 AQ-associated non-AQ-associated exhibited most prominent differences. RT-PCR confirmed significant increase TP63 Serpine1 mRNA expression OSCC relative non-malignant matched tissue. A immunoreactivity observed from tissue OSCC. However, there no correlation between frequent loss Caspase 8 decrease expression. These data demonstrate that profiling can be useful analysing pathogenetic events involved genesis or progression
