摘要: Human embryonic stem cells (hESCs) represent a potential source of blood for transfusion therapies and promising tool studying the ontogeny hematopoiesis. Moreover, human-induced pluripotent (hiPSCs), recently established by defined reprogramming factors expressed in somatic cells, further generation hematopoietic cells. When undifferentiated hESCs or hiPSCs are cultured on either mesenchymal C3H10T1/2 OP-9 stromal they can be differentiated into niche that concentrates progenitors, which we named "embryonic cell-derived sacs" (ES-sacs). We have optimized vitro culture condition obtaining mature megakaryocytes derived from progenitors within ES-sacs, then able to release platelets. These vitro-generated platelets display integrin activation capability, indicating normal hemostatic function. This novel protocol thus provides means generating as well hiPSCs, study human thrombopoiesis also disease conditions using patient-specific hiPSCs.
