Influence of poly(DL-lactide) nanocapsules on the biliary clearance and enterohepatic circulation of indomethacin in the rabbit.

摘要: Following intravenous administration, the uptake of colloidal drug carriers by cells mononuclear phagocyte system, mainly Kuppfer cells, may concentrate an encapsulated close to liver parenchymal and facilitate its biliary excretion enterohepatic circulation. To test this hypothesis indomethacin was administered (10 mg/kg) in four groups 10 rabbits each infusion at a constant rate over 2 hr, either free form (aqueous solution) or as nanocapsules prepared from preformed poly(DL-lactide). Unchanged assayed plasma two control (sham-operated) both bile bile-cannulated groups. Pharmacokinetic analysis led conclusion that macrophages reduces concentration enhancing total clearance. This enhancement due increase clearance, result parallel increases drug. It also demonstrated enhance circulation indomethacin.
