Allele-specific FKBP5 DNA demethylation mediates gene-childhood trauma interactions

摘要: Although the fact that genetic predisposition and environmental exposures interact to shape development function of human brain and, ultimately, risk psychiatric disorders has drawn wide interest, corresponding molecular mechanisms have not yet been elucidated. We found a functional polymorphism altering chromatin interaction between transcription start site long-range enhancers in FK506 binding protein 5 (FKBP5) gene, an important regulator stress hormone system, increased developing stress-related adulthood by allele-specific, childhood trauma-dependent DNA demethylation glucocorticoid response elements FKBP5. This was linked stress-dependent gene followed long-term dysregulation system global effect on immune cells areas associated with regulation. identification genotype-directed reactivity will be useful for designing more effective treatment strategies disorders.
