摘要: We use terahertz (THz) spectroscopy as a biomaterials characterization tool. Previously we have shown strong contrast between the THz dielectric response for wild type (WT) and D96N mutant of bacteriorhodopsin. In those studies observed large increase in absorbance WT with excitation to thermally captured photo-intermediates whereas no such was D96N. These results suggest that is sensitive structural changes relative flexibility biomolecules. However photo-intermediate populations samples were not equivalent measurements. While had relaxed (bR), M P state intermediates present, only bR states. Here present measurements function at room temperature. The constant intermediate states up 23% 30% state. verify there fundamental difference conformational dynamics measured by single residue mutation.
