摘要: In the present work, to analyze heterogeneity of tumor-specific cytotoxic immune response, a large number T cell clones were generated from infiltrate tumor-proximal invaded lymph node, and two kinds melanoma-specific CD8+ CTL derived. The majority (about hundred) are characterized by specific lysis autologous tumor lines. Among 34 latter clones, HLA-A2 molecule MART-1(27-35) peptide have been shown play predominant role in recognition. However, no significant amplification at site was observed for 3 these CTL. other kind (1 oligoclonal 2 clonal lines) did not lyse melanoma lines but lysed "fresh" cells MHC class I-dependent manner. Functional analysis different that they NK targets, peripheral monocytes, activated cells, transformed B or any few allogeneic cultured uncultured we tested. TCR repertoire has one significantly detectable "in situ" among tumor-infiltrating lymphocytes, while PBMC. Such which could picked out through conventional screening procedures using lines, potentially rejection. These results suggest response analyzed toward does totally reflect situ status.
