摘要: The production of viable biological heart valves is central interest in tissue engineering (TE). aim this study was to generate decellularized with an intact ultra-structure and repopulate these endothelial cells (EC) under simulated physiological conditions. Decellularization ovine pulmonary valve conduits performed agitation detergents followed by six wash cycles. Viability EC cultures exposed washing solution served prove efficiency washing. Resulting scaffolds were free preserved extracellular matrix. Biomechanical standard tension tests demonstrated comparable parameters native tissue. Luminal surfaces valvular grafts seeded jugular vein dynamic bioreactors. After rolling culture for 48 h, pulsatile medium circulation a flow 0.1 L/min started. incremented 0.3 L/min/day up 2.0 (cycle rate: 60 beats/min), while pH, pO2, pCO2, lactate glucose maintained at constant levels. 7 days, monolayer covered the inner surface, which expressed vWF, indicating origin. A complete endothelialization detergent scaffold can be achieved conditions using bioreactor system, allows continuous control environment.
