摘要: PKZ is a fish-specific protein kinase containing Zα domains. known to induce apoptosis through phosphorylating eukaryotic initiation factor 2α (eIF2α) in the same way as double-stranded RNA-dependent (PKR), but its exact role detecting pathogens remains be fully elucidated. Herein, we have found that acts DNA sensor by initiating IFN expression IRF3- or ISGF3-like mediated pathways. The pattern of similar those innate immunity mediators stimulated poly (dA:dT) and (dG:dC). DNA-PKZ interaction can enhance phosphorylation dimerization vitro. These findings indicate participates cytoplasmic DNA-mediated signaling. Subcellular localization assays also shown located cytoplasm, which suggests PRR. Meanwhile, co-IP separately interact with IRF3, STING, ZDHHC1, eIF2α, IRF9, STAT2. Further investigations revealed activate IRF3 STAT2; IRF3-dependent dependent are critical for PKZ-induced expression. results demonstrate special pattern-recognition receptor, trigger immune responses IRF3-mediated pathways fish.
