Retinoid Compounds Activate and Induce Hepatic Glycine N-Methyltransferase in Rats

摘要: Glycine N-methyltransferase (GNMT) functions to regulate S-adenosylmethionine (SAM) levels and the ratio of SAM/S-adenosylhomocysteine (SAH). SAM is a universal methyl group donor up-regulation GNMT may lead wastage groups required for transmethylation reactions. Previously, we demonstrated that dietary treatment rats with 13-cis-retinoic acid (CRA) decreased hepatic concentration SAH ratio. Here, examined ability CRA, as well all-trans-retinoic (ATRA), potential basis our earlier observations. Rats were fed either control (10% casein + 0.3% L-methionine) diet or supplemented L-methionine (10 g/kg diet). from each orally given ATRA, CRA (both at 30 micromol/kg body), vehicle daily 7 d. For rats, administration both ATRA elevated activity 49% 34%, respectively, compared group. Similar results exhibited by methionine-supplemented diet. Moreover, retinoid-induced elevations in enzyme reflected abundance protein. To knowledge, this first report nutritional compound induces transcriptional and/or translational level.
