摘要: Peripheral blood monocytes are common precursor cells of dendritic (DCs) and macrophages. We have searched for factors with the potential to regulate differentiation DCs When CD14+ cultured granulocyte-macrophage colony-stimulating factor (GM-CSF) interleukin (IL) 4, CD14+CD1a- population, which consists macrophages, was found in serum-containing cultures but not serum-free cultures. Addition IL-6 receptor-neutralizing monoclonal antibody (mAb) or gp130-neutralizing mAb resulted a decreased population cells. An increase reduction CD14-CD1a+ observed addition cultures, whereas IL-11, leukaemia inhibitory factor, oncostatin M macrophage did affect presence GM-CSF plus IL-4. This effect blocked by tumour necrosis alpha (TNF-alpha), lipopolysaccharide (LPS), IL-1beta, CD40 ligand (CD40L) transforming growth beta1 (TGF-beta1). Among these factors, TNF-alpha most potent interfering action IL-6. These results suggest that inhibits promoting their toward is modulated such as TNF-alpha, LPS, CD40L TGF-beta1.
