Effects of miR - 19b Overexpression on Proliferation, Differentiation, Apoptosis and Wnt/β-Catenin Signaling Pathway in P19 Cell Model of Cardiac Differentiation In Vitro

摘要: MicroRNA (miR)-19b is part of the miR-17–92 cluster associated with cardiac development. Here, we investigated effects overexpressing miR-19b on proliferation, differentiation, apoptosis, and regulation Wnt/β-catenin signaling pathway in multipotent murine P19 cell line that can be induced to undergo cardiogenesis. cells were transfected plasmid or empty vector, overexpression was verified by Quantitative Real-Time PCR (qPCR). The vector control stable lines selected using Blasticidin S HCl, their cycle, apoptosis levels analyzed Cell Counting Kit-8 flow cytometry. differentiation markers, apoptosis-related genes (bax, bcl-2), pathway-related detected qPCR, corresponding proteins Western blot. Expression Wnt marker also immunofluorescence. Morphological changes observed electron microscopy Hoechst staining. On basis these results, demonstrated promoted proliferation but inhibited cells; β-catenin expressions decreased, while GSK3β increased overexpression. Overexpression activation cells, which may regulate cardiomyocyte differentiation. Our findings bring new insights into mechanisms underlying diseases suggest a potential therapeutic target for cardiovascular diseases.
