摘要: Detergents have been used to enhance the solubility of hydrophobic biomolecules for decades. Despite widespread use detergents in biochemistry, presence these molecules often complicates further analysis by mass spectrometry. This study presents a solution this problem outlining method utilizing novel cleavable detergent, 3-[3-(1,1-bisalkyloxyethyl)pyridin-1-yl]propane-1-sulfonate (PPS). detergent can be extract protein contained within interior cell disrupting membranes. Once proteins are free from cell, PPS also assists solubilization shielding regions newly extracted unfavorable interactions with water. The added advantage over conventional such as sodium dodecyl sulfate or n-octylglucoside is that properties interfere MALDI spectrometry eliminated prior analysis. was found improve sensitivity analyses both soluble and membrane without degrading spectral quality. virtues strategy were applied whole extracts. Analysis extracts resulted an overall increase number peaks observed signal intensity.
