Contractility of myofibrils from the heart and diaphragm muscles measured with atomic force cantilevers: Effects of heart-specific deletion of arginyl-tRNA–protein transferase

摘要: Abstract Background Contractile properties of myofibrils from the myocardium and diaphragm in chronic heart failure are not well understood. We investigated a knockout (KO) mouse model with cardiac-specific deletion arginyl-tRNA–protein transferase (α-MHCAte1), which presents dilated cardiomyopathy failure. Objective The aim this study was to test hypothesis that α-MHCAte1 mice is associated abnormal contractile diaphragm. Methods used newly developed system atomic force cantilevers (AFC) compare age-matched wild type (WT). Myofibrils were attached AFC for measurements during activation/deactivation cycles at different sarcomere lengths. Results In heart, presented reduced full activation (89±9nN/μm 2 ) when compared WT (132±11nN/μm ), decrease influenced by length. These similar kinetics development (K act redevelopment tr relaxation rel ). diaphragm, an increased (209±31nN/μm (123±20nN/μm Diaphragm K , but faster (6.11±0.41s −1 vs 4.63±0.41s Conclusion Contrary our working hypothesis, produced WT. results suggest potential compensatory mechanism works under loading conditions model.
