摘要: Alzheimer's disease (AD)—the most common type of dementia among the elderly—represents one challenging and urgent medical mysteries affecting our aging population. Although dominant inherited mutation in genes involved amyloid metabolism can elicit familial AD, overwhelming majority AD cases, dubbed sporadic do not display this Mendelian inheritance pattern. Apolipoprotein E (APOE), main lipid carrier protein central nervous system, is only gene that has been robustly consistently associated with risk. The purpose current paper thus to highlight pleiotropic roles structure-function relationship APOE stimulate both functional characterization identification novel homeostasis-related molecular targets AD.
