摘要: We sought to determine the genetic basis of expression ubiquitous (metabolic) urease soybean. This isozyme is termed metabolic because its loss, in eu4/eu4 mutants, leads accumulation urea, whereas loss embryo-specific does not. The eu4 lesion eliminated vegetative and embryonic tissues. RFLP analysis placed clone LC4 near, or within, Eu4 locus. Sequence comparison proteins (ubiquitous embryo-specific) clones (LC4 LS1) indicated that LS1 encode ureases, respectively. That transcribed into poly (A)+ RNA all tissues was by amplification transcript an LC4-specific PCR primer. (The LS1-specific primer, on other hand, amplified poly(A)+ only from developing embryos expressing urease.) These observations are consistent with being structural gene contained clone. In agreement this notion, mutant phenotype callus partially corrected introduced particle bombardment.
