摘要: Leginsulin, a peptide made up of 37 amino acids, is homologous to pea (Pisum sativum L.) albumin (PA1b) and belongs the cysteine-knot family. Even though physiological function threedimensional structure leginsulin have been explored, little known about its expression, accumulation, distribution among soybean [Glycine max (L.) Merr.] accessions. An antibody generated against was used screen diverse array accessions from USDA Soybean Germplasm Collection identify that are enriched in leginsulin. Analysis 50% isopropanol-soluble proteins 485 lines by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDSPAGE) western blot analysis enabled identifi cation 75 signifi cantly accumulated Remarkably, all these with exception two had their origin Japan, Korea, or China. Leginsulin barely detected most commercial North American cultivars this study. Western revealed low accumulation embryonic axis but not cotyledons cultivar Williams 82 while Chinese PI 458249 abundantly present both types tissue. Examination genome sequence presence genes (Gm13 g26330 [leginsulin 1] Gm13 g26340 2]) on chromosome 13. We cloned found them be highly similar 82. Northern indicated messenger RNA (mRNA) abundant cotyledon. In contrast, mRNA 458249, cotyledons.
